JA: Are you fixing your Highlander Hybrid yourself? What have you tried so far?
Customer: removed panel to access lock mechanism, attempted to manually unlock

have you tried resting in case it was a electrical glitch ,did you remove rear seat to access the rear trunk actuators

Master reset Procedure

1- Disconect Battery Cables ( Negative & Positive Cables )you can use any electrical wire as an extension if battery cables don't reach .

2- Hold cables Together for 60 secs ,then let go

3- Wait 15 minutes

4- Make sure all doors are closed

5- Reconnect battery Cables

6- Lock & unlock the vehicles using the key from driver door side (if you have a key fob with no blade key, just lock & unlock using the key fob buttons)

7- Enter the vehicle and attempt to start

then you need to take the rear trim apart from the lift gate going thru from inside the car to get the latch and actuator

Hello and welcome to JustAnswer!

If you've gained access to the latch, found the linkage is intact and you're unable to get it to open moving the linkage manually by hand then the problem is with the latch itself, provided there is no damage to the body or liftgate and there is nothing foreign closed in the liftgate opening. If the latch has failed then it will need to be cut/broken/dismantled until the pawl can be moved manually to open the liftgate and then replace the latch.
