Paracetamol or acetaminophen, as it is referred to in the states, has no literature to support the development of tolerance. There are other pain medications, primarily narcotics, that tolerance can be built up with prolonged use.
As for will it stop working, that depends. The maximum total daily dose should never exceed 4,000 mg per day, and regular use of this dose is not recommended. If paracetamol at total daily doses of 4,000 mg per day (i.e.: 1,000 mg every 6 hours) is not effective, I'd recommend switching to a stronger non-prescription medication. Those may include, but are not limited to: ***** ***** excedrin, etc... Many patients have headaches that require stronger medications to treat. Once you've exhausted your non-prescription options, I would recommend seeing your doctor for a prescription medication. If you are getting these headaches on a frequent enough basis, you might want to consider getting evaluated for other possible causes.
