I'm assuming the power locks work ok.
If the issue is only with the lock button on the fob and the unlock button does work then the problem is almost surely the fob itself.
I the power locks do work but the keyless entry doesn't then again the fob is the most likely cause.
What should be done is test the fob's output. This can be done by a dealer or may independent shops but most big chain parts stores such as Autozone will test fobs for free. If the battery is good and the fob is not transmitting when the lock button is pressed then it has failed.
