the door behind the driver seat is closed and I am unable to unlock it and unable to open it also
JA: Is this your first time dealing with this issue?
Customer: I had many issues of not unlocking but I was always able to at lease unlock it manually by the inside.
JA: What is the model/year of your Lexus?
Customer: RX350 2010
JA: Are you fixing your vehicle yourself? What have you tried so far?
Customer: for the door issues not much yet. I have removed the door pannel and put it back in place this morning in order to get the part number of the door actuator motor.
JA: Anything else you want the mechanic to know before I connect you?
Customer: I don't think so

Ok, you will have to remove the interior door handle and pull the linkage manually

I tried but the handle plastic panel seems to be
glued to the door pannel. Is the only way to do it is to break that panel or is there a way by the window?

The panel does not need to be broken. After you remove the cover plate for the interior door handle, you can taeke it off by removing the screws behind the plate.

I've tried very hard but it is stick on the 4 corners. It is kind of a rectangular shape. my tool can slide only on the two longer side (top and bottom). Is there a plan B?

the screw in the middle (behind the plastic pannel) is removed

Unfortunately that is the only way to remove it with the door closed.
