You are going to want to remove the pepper from the plant when it is ripened. When you try to pull the pepper, it should pull easily as this shows it is ripe. If it does not pull easily, it is not ready to take off the plant.

Also, depending on your variety of chili pepper, it might get a brown stripe down it - this is also an indicator of being ripe and ready.
