I normally use anti-sieze on all the plugs.

To remove the number 5 cylinder ignition coil, remove the evaporative emission (EVAP) canister purge tube.

Should I remove wire connection from coil packs before removing or doesn't it matter?

They normally will have enough slack to just lay them aside but either way won't really matter.
