Hi,
If this happened quickly, it most likely is a bee sting or spider bite.
Benadryl is OK, but does not work as well in dogs as people.
If her neck is really swollen, and she is panting from the swelling, she needs to see the vet for a quick dose of steroids to help bring the area under control quickly. There is nothing over the counter that will help in these circumstances.
