Question

I currently work for one department in an engineering company. I am being told that I have to take a job in another division that requires an immediate 6 month assignment 500 miles from my home. If I say no am I saying that I quit or does the company have to fire me?

That depends on your specific contract with the company - if contract terms address your relocation and termination policy.

If you are an employee at-will - either - you or your employer may terminate employment at any time.

Your refuse to relocate not necessary means you are fired, but the company MAY fire you because of that. In this case you should be eligible for unemployment compensation.
