It sounds like you have a stoppage. Most likely in the toilet bowl but it's also possible (based on your description) that the sewer is plugged. The best option to clear a stoppage in the toilet is a specially designed snake called a closet/toilet auger (see attached pic). If that doesn't correct the problem then the sewer will need to be snaked out with a large motorized snake.
