I was recently diagnosed with Cushing disease. I have an adrenal adenoma measuring 3.4. , facial hair, painful hands and hips, buffalo hump, lots of belly fat, no red stretchmarks. I took one prednisone 40mG tablet on Monday and they did a cortisol test on Thursday that was 2.9. Can this be a false positive

Yes. YOu have all of the signs and symptoms of cushing's disease.
If you took a prednisone tablet, it can affect the cortisol level
Patients taking prednisone may have falsely increased cortisol levels because prednisone is converted to prednisolone after ingestion and prednisolone has a 41% cross-reactivity.
