Garmin741xs... I have new software update but can not load because my touch screen is just not reacting. Garmin says I need a touch screen calibration. Can you help me?

While it is not normally needed to be done, you can calibrate the screen by going into settings> system> display> calibrate> Display. then follow the on screen instructions to get the calibration complete.

We tried the instructions you suggested from Garmin. Touch screen will not react to any touch button prompts except the "home" button and then it will just not allow any further propts???
Any other suggestions other than call Garmin on Monday?

This is really sounding like you have a bad screen, but truly only Garmin can make that call. Most likely they will want you to sent the unit to them so that they can repair it for you.

How old is the unit?

Sorry, I was looking up Garmin's warranty policy. It looks to only be a year. I did find where on their site, they would overhaul your unit for $300.
