Create a new image on the drive that fills the disk using Disk Utilities - you can use 128 or 256 bit encryption. You can reformat the drive, etc. as desired, first.

When you want to access the image, dbl-click on the sparsebundle on the disk, and the OS will ask you for a pwd, open the sparsebundle, and mount it as another drive.

I know it seems like a roundabout method, but it is really the only way to make sure nobody can access it except you.
