I am working on a 2000 Lexus GS300. I did a complete cylinder head job which was resurfaced .003 and complete valve job including the valve lash shim style. I am positive that I never touched the lifter buckets and even marked their position just in case.
New plugs, radiator, thermostat, coolant temp sensor, timing belt , tensioner, and pulley. Attempted to start it after assembling. Car sat a total of 5 weeks. Cranks but no start. #1 & 6 plugs wet and those cylinders have good compression.. Fuel pressure good,
rechecked timing marks crank pulley and camshaft pulleys. Spark check #1 only one flicker, attempts to run but doesn't,maybe 5 seconds, then has a soft backfire thru the intake. No codes. Any ideas?

What is compression on each cylinder?

Do a spark check on every cylinder.

Check for injector pulse on each cylinder
