If a former coworker finds out that I am applying at his current workplace goes to his employer and tells them I'm a bad person lazy ect.. Could I sue for defamation?

Yes, if this person told lies about you then you can sue for damage to your reputation.

As a first step you should consult with a lawyer face to face with a view to retaining the lawyer to send this person a letter demanding a written retraction and a formal apology and threatening a lawsuit.

If that does not work the next step would be to sue.
