Cyinder #5 is on the side of the engine closest to the windshield, 3rd one from the passenger's side (see image below). There are a few things that can cause a single-cylinder misfire: a faulty spark plug, a faulty ignition coil, a faulty fuel injector, an air intake leak around the affected cylinder, low cylinder comrpession, etc. The most common will be a faulty plug or ignition coil, so this would be the best place to start.
